DESCRIPTION:Professor Ying DiaoDepartment of Chemical and Biomolecular EngineeringUniversity of Illinois\, Urbana-Champaign \n\n\n\n \n\n\n\nAI-guided Closed-loop Learning for Functional Molecule Discovery\n\n\n\n\n\n\n\nClosed-loop discovery of functional molecules represents a new paradigm to new materials discovery. However\, so far\, AI-driven closed-loop discovery approaches have rarely yielded new chemical insight. I am leading a team to address this challenge under the Molecule Maker Lab Institute\, one of the first AI Institutes in US. We connect autonomous synthesis\, autonomous device printing and testing\, and machine learning in a closed loop to drive molecular design of light harvesting small molecules towards high photostability. We combined blind Bayesian Optimization (BO) with physics-based ML models to expand the classical “closed loop optimization” method for a more informative “Closed-Loop Transfer” (CLT) paradigm capable of making scientific discoveries We uncovered a first principles understanding of the molecular determinants of photostability in which the triplet excited state manifold plays a critical role against literature findings. We further applied this strategy to understanding molecular design rules underpinning chiral emergence of conjugated polymers. Such understanding laid the foundation to developing next generation chiral electronics spanning solar cells\, conductors\, spintronics and beyond. At present\, we are working to partner Large Language Model with our closed-loop learning approach towards developing a chemical language model for organic electronics.

MillerSenior Research ScientistSchlumberger-Doll Research Center \n\n\n\nAn industrial perspective on membrane research\n\n\n\n\n\n\n\nPolymeric membranes are an energy-efficient alternative to thermally driven separation processes. Natural gas processing is one of the largest industrial markets for gas separation membranes. Virtually all natural gas must be treated to remove carbon dioxide (CO2) and hydrogen sulfide (H2S) before entering a pipeline\, as these acid gases can corrode the pipeline in the presence of water. Cellulose triacetate (CTA) asymmetric hollow fiber membranes are effective for removal of acid gases from natural gas\, although they are subject to plasticization and limitations on process conditions. While thousands of experimental materials—some with remarkable separation characteristics—have been proposed as alternatives\, few have reached commercialization and legacy materials such as CTA continue to dominate. This presentation will provide an overview of ongoing industrial membrane research for natural gas processing. Current needs in membrane material science will be discussed. Because membrane modules operate within an ecosystem of other system components\, the impact of membrane material properties will be contextualized among cost drivers in natural gas processing. Finally\, the role of modeling will be explored\, including prediction of module performance from laboratory-scale transport measurements\, which can then be used to direct material discovery efforts to shorten time to commercialization.

DESCRIPTION:Susan DanielDistinguished ProfessorDepartment of Chemical and Biomolecular EngineeringCornell University \n\n\n\nToward objective pain monitoring and treatment: Cell-free biosensors based on nociceptive ion channels\n\n\n\n\n\n\n\nEffectively monitoring and managing pain remain major challenges in the clinic and constitute active areas of research today. Given the subjective nature of the pain response and the ongoing opioid crisis\, there is a critical need to both identify new non-opioid drug targets for chronic pain management and provide more objective ways to measure pain. Using pain receptors themselves to monitor biomarkers of pain and screen potential channel blockers remains challenging\, with current drug screening protocols lacking quantitative and direct measures of receptor function and dysfunction. To improve the treatment and measurement of pain\, we describe here the development of prototypical pain biosensors. We utilize in vitro transcription and translation cell-free methods to rapidly express and integrate two distinct and pharmacologically relevant types of pain receptors into membrane-based sensing platforms: the Transient Receptor Potential Vanilloid 1 (TRPV1) channel and pro-algesic P2X purinergic receptors (P2X2)\, avoiding all the downsides of cell-based expression\, isolation\, and reconstitution into membranes and sensor platforms. Because these receptors are ligand-gated ion channels activated by different stimuli\, we then assess both channels’ responsiveness using electrical or optical methods to monitor changes in ionic flux across the membranes. \n\n\n\nIn our first system\, we show that devices containing P2X2 receptors show an increase in ionic flux following ATP administration\, indicating ligand-gated channel opening. This ion flux is abolished in the presence of a known P2X2 inhibitor\, but not the P2X7 inhibitor\, demonstrating the screening potential of this platform to identify new subtype specific therapeutics for pain management. Using the same in vitro expression system\, we then cell-free synthesized and integrated the human nociceptor TRPV1 channels into membrane vesicles containing a fluorescence-based ion flux reporter. In these vesicles TRPV1 channel opening could be stimulated by two distinct pain-causing stimuli – heat and capsaicin\, showing that in our minimal system\, this receptor subtype is responsive to distinct channel activators. The modular nature of these two platforms enables isolation and assessment of individual pain receptors in a way that is not possible using native cells in a convenient\, tailorable device compatible with rapid\, high throughput screening. Both electrical and optical platforms\, which contain cell free synthesized pain receptors\, provide new ways to study receptor function and advance our detection and treatment of pain.

Lapidus ProfessorDepartment of Chemical and Biomolecular EngineeringUniversity of Massachusetts Amherst \n\n\n\nPolyelectrolyte Complex Materials\n\n\n\n\n\n\n\nElectrostatic interactions can be used in the self-assembly of a wide range of responsive\, bioinspired soft materials ranging from dehydrated thin films\, fibers\, and bulk solids to dense\, polymer-rich liquid complex coacervates\, as well as more complex hierarchical structures such as micelles and hydrogels. This responsiveness can include swelling and dissolution or solidification\, which can be harnessed to facilitate encapsulation and the subsequent fabrication of functional materials. We draw inspiration from biomolecular condensates\, which utilize liquid-liquid phase separation to create transient compartments in cells. These condensates form due to weak\, multivalent interactions involving intrinsically disordered proteins\, and have been shown to enable the selective uptake of specific proteins\, nucleic acids\, and small molecule drugs. We study how the patterning or presentation of charges and other chemical functionalities can modulate the potential for liquid-liquid phase separation via complex coacervation. We further examine how the distribution of charge on proteins and viruses can be used to facilitate selective uptake into coacervate phases\, and stabilization against denaturation. This molecular-level understanding of polyelectrolyte complexation is further enhanced by detailed dynamic mechanical and thermodynamic examinations of the molecular nature of the various material transitions present in these systems. Our experimental efforts are supported by the parallel development of computational approaches for modeling and predicting the phase behavior of patterned polymeric materials.  \n\n\n\nOur goal is to establish molecular-level design rules to facilitate the tailored creation of materials based on polyelectrolyte complexation that can both illuminate self-assembly phenomena found in nature and find utility across a wide range of real-world applications.

DESCRIPTION:Zhen MaAssociate ProfessorDepartment of Biomedial and Chemical EngineeringSyracuse University \n\n\n\nAt the crossroads of new approach methodologies and artificial intelligence\n\n\n\nZhen Ma Portrait\n\n\n\nThe convergence of artificial intelligence (AI) and human induced pluripotent stem cell (hiPSC) platforms is redefining in vitro biological models. Advances in hiPSC-derived three-dimensional cardiac organoids\, engineered heart tissues\, and heart-on-chip systems have improved physiological relevance but generate high-dimensional\, multimodal datasets that exceed the capacity of traditional analytical approaches. In this seminar\, I will discuss how emerging computational approaches\, including machine learning\, deep learning\, and generative AI\, can unlock the full potential of engineered cardiac models. By integrating AI with in vitro cardiac models\, we demonstrate how data-driven approaches can enable scalable quality control\, reveal subtle disease phenotypes\, and guide rational model design. Together\, these advances position AI as a powerful co-pilot in cardiac research\, accelerating cardiotoxicity screening\, improving disease modeling\, and informing the next generation of intelligent cardiac organoids.
